What is Proline? Proline is a non-essential amino acid — meaning your body can produce it, but many people don't make or get enough to meet demands during injury, aging, or high physical stress. It is uniquely structured compared to other amino acids, forming a ring shape that gives collagen its tight, coiled stability. Without adequate proline, collagen production slows and connective tissue integrity suffers. How does Proline work? Proline is one of the two key amino acids (alongside hydroxyproline) that make up roughly 25% of all collagen. Your body incorporates proline directly into collagen strands, and then converts some of it to hydroxyproline — a step that requires vitamin C. This gives collagen its triple-helix structure, the tight coil that makes tendons, skin, and cartilage strong and elastic. Proline also plays a supporting role in wound healing by stimulating fibroblasts, the cells that lay down new tissue. What forms does Proline come in? Available as standalone L-Proline powder or capsules, or obtained indirectly through hydrolyzed collagen peptide supplements. Powder form allows flexible dosing and is often combined with vitamin C for enhanced collagen synthesis. Collagen peptides (types I, II, III) provide proline alongside hydroxyproline and glycine in a pre-digested, highly absorbable form. Where it comes from: Proline was first isolated in 1900 by Richard Willstätter. Its central role in collagen structure has been well understood since the mid-20th century, and it became a focus of nutritional and sports medicine research as interest in collagen synthesis and connective tissue repair grew in the late 1990s and 2000s.

Helps

What does Proline help with? Skin aging and loss of firmness: Collagen makes up most of your skin's structural layer, and proline is its main building block. As proline availability declines with age or poor diet, collagen production slows, leading to thinner, looser skin. Supplementing proline — especially alongside vitamin C and glycine — helps your skin rebuild its collagen scaffolding, improving firmness and reducing fine lines over time. Joint pain and cartilage wear: Cartilage is almost entirely made of collagen, and proline is essential to keeping it strong and resilient. When cartilage breaks down faster than it's rebuilt — as in osteoarthritis or heavy physical training — extra proline helps tip the balance back toward repair. Many joint supplement studies use collagen peptides, which deliver proline directly to cartilage-building cells. Wound healing and tissue repair: When you have a wound, your body rapidly assembles new collagen to close it — a process that demands a surge of proline. Research shows that adequate proline availability speeds wound closure and improves scar quality. This makes it especially relevant after surgery, burns, or injury. Leaky gut and intestinal lining: The lining of your gut is reinforced by collagen, and proline helps maintain its tight junctions — the seals between intestinal cells that keep bacteria and toxins from leaking through. A proline-rich diet or targeted supplementation may help support gut barrier integrity, particularly in people with inflammatory bowel conditions or intestinal permeability. Tendon and ligament injuries: Tendons and ligaments are dense collagen structures with very low blood flow, meaning they heal slowly. Providing proline — ideally with vitamin C before exercise or physical therapy — gives these tissues the raw materials they need to lay down new, stronger collagen during the repair window after activity. Post-surgical recovery: Surgical wounds require rapid collagen synthesis to close properly, and the body's demand for proline spikes during this period. Clinical nutrition protocols for surgical patients often include amino acid support, and adequate proline status has been linked to better wound closure speed and reduced complication rates. Stretch marks and skin damage: Stretch marks form when skin is stretched faster than collagen can be produced, causing the dermis to tear. While no supplement reverses existing stretch marks completely, supporting collagen synthesis with proline may help improve skin resilience and reduce the depth and visibility of new marks over time. Exercise-related connective tissue stress: High-load exercise stresses tendons, ligaments, and joint cartilage in ways that muscles often recover from faster. Athletes benefit from targeted collagen and proline support, particularly when timed around training sessions to take advantage of the post-exercise tissue-building window. Bone density support: Bone isn't just calcium — about a third of bone volume is collagen, which acts as the flexible framework that calcium crystals attach to. Adequate proline supports the collagen matrix that makes bone tough and fracture-resistant, complementing calcium and vitamin D intake.

This app is so cool! I've always wanted there to be a hub for all NATURAL remedies and preventative options but l've always had to do loads of digging to find the info I wanted for different natural products. HealisticMD has all the info I need in one place. It gave me various natural supplemental options for a condition I looked up, including what each remedy does and how much is usually recommended to take. It also gave me exact products I could buy for the problem I had looked up. It makes my process SO MUCH easier when I need to find non-pharmaceutical solutions to my needs.

There is a wealth of information on the app! I recently had bloodwork done and found I was deficient in a handful of vitamins and minerals (Vitamin B12, Vitamin D, Iron, etc.) HealisticMD made it simple for me to find out what foods to eat/ supplements I needed to improve these deficiencies. It gives you so many ideas on prevention, natural health tips, where to buy the vitamins, when to take them, why, etc. I love how easy the app is to use too. I am currently working on getting pregnant. I was pleasantly surprised to find compelling data along with supplement suggestions to aid in the process, some of which my fertility doctor wasn’t even aware of. Highly recommend downloading, after all health is one our greatest gifts!
